基于洛美沙星免疫原的喹诺酮药物广谱特异性抗体制备的研究

Preparation of broad-specific antibody to fluoquinolone drugs based on lomefloxacin immunogen

  • 摘要: 以洛美沙星为半抗原,采用碳二亚胺法和活泼脂法将洛美沙星分别偶联于牛血清白蛋白(BSA)和卵清白蛋白(OVA)制备出免疫原和包被抗原,免疫新西兰大白兔获得了抗体。经间接竞争酶联免疫吸附分析方法(icELISA)鉴定,首次基于洛美沙星一种半抗原获得了能够识别至少11种喹诺酮药物的抗体,且检测限低于10ng/mL,灵敏度满足相关标准检测要求。这对进一步开发喹诺酮类药物多残留检测试剂盒具有重要意义。 

     

    Abstract: Using lomefloxacin as hapten, the immunogen was prepared with water soluble carbodiimide (EDC) method, and the coating agent was synthesized with active ester method.The polyclonal antibody was obtained successfully after immunization.Characterized by indirect competitive enzyme linked immunosorbent assay (icELISA) , the antibody exhibited strong broad-specificity, 11 fluoroquinolone drugs could be recognized with a limit of detection lower than 10ng/mL, which could meet the requirement of related international organization.This study would benefit to the immunoassay development for the simultaneous detection of many fluoroquinolone drugs.
