响应面法优化超声提取甘青铁线莲总皂苷

Optimization ultrasonic extraction process of total saponins from Clematis tangutica (Maxim.) Korsh by response surface methodology

  • 摘要: 利用单因素实验及响应面法优化确定甘青铁线莲总皂苷的提取参数条件。通过单因素实验确定乙醇浓度、超声时间、料液比三因素的最佳水平。以甘青铁线莲总皂苷提取得率为响应值进行Box-Behnken中心组合实验设计,建立铁线莲总皂苷提取得率的二次回归方程,得到优化组合条件。响应面法分析结果表明,当乙醇体积分数为80%,超声提取时间50min,料液比1:30时,铁线莲总皂苷提取得率最大为15.78mg/g,接近于模型预测值16.19mg/g。 

     

    Abstract: Single-factor tests and response surface method (RSM) were used to optimize the extraction process of total saponins in Clematis tangutica (Maxim.) Korsh.In order to obtain the maximum yield of the extraction of total saponins, three main factors including alcohol concentration, ultrasonic extraction time and ratio of raw material to alcohol were determined with the Box-Behnken design and established a quadric regression equation to predict the yield of total saponins.Results showed that the optimum extraction conditions were as follows:alcohol concentration 80%, ultrasonic extraction time 50min, and ratio of raw material to alcohol 1:30.Under the conditions, the optimum extraction yield of total saponins was 15.78mg/g, which was well matched with the predictive extraction yield 16.19mg/g.
