高效液相色谱-示差折光法测定甜菊糖中莱鲍迪苷A的含量

Separation and identification of rebaudioside A in stevia by high performance liquid chromatography-refractive index detector

  • 摘要: 建立了高效液相色谱-示差折光方法来测定甜菊糖中莱鲍迪苷A含量的实验方法。色谱柱采用WatersNH2柱(250mm×4.6mm,5μm),以乙腈∶水=80∶20为流动相,采用示差折光检测器测定甜菊糖中莱鲍迪苷A的含量。莱鲍迪苷A的线性范围为0.1~1.0mg/mL,检出限为0.03mg/mL,平均加标回收率为98.37%,相对标准偏差0.88%。该方法简单、快速、准确、重现性好,适用于甜菊糖中莱鲍迪苷A的定量分析。 

     

    Abstract: A high performance liquid chromatography method for separation and determination of rebaudioside A (RA) in stevia was established. The analytical column was Waters NH 2 (250mm ×4.6mm, 5μm) , the mobile phase was acentonitrile -water (8020 , V/V) , the samples were determined by an refractive index detector detector. The calibration curves had good linearity in the range of 0.1~1.0mg/mL for RA, the detection limit for RA was 0.03mg/mL, the average recoveries of RA were 98.37% (n=6, RSD=0.88%) . The method was convenient, rapid and effective for qualitatively analysis of RA in stevia.
