花生发根培养条件的优化

Optimization of components in culture medium for hairy roots of peanut (Arachis hypogaea L.)

  • 摘要: 利用单因素实验和正交设计研究了培养基中激素、碳源、氮源、磷酸盐、初始pH等对鲁花11号花生发根生长的影响。结果表明,花生发根的最适基本液体培养基为无激素的MS培养基;培养基中加入NAA、6-BA、IBA等三种激素,在不同程度上抑制发根的生长;花生发根培养最适条件为:碳源为1.5%蔗糖、氮源含量与标准MS培养基相同、磷酸盐为3.75mmol/L、初始pH为6.1。100μmol/L水杨酸(SA)可以提高花生发根白藜芦醇苷合成分泌以及白藜芦醇的合成。 

     

    Abstract: Effects of phytohormone, carbon source, nitrogen source, phosphate, initial pH value of medium on growth of hairy roots of peanut variety Luhua No.11 were studied through optimization of culture medium using single factor and orthogonal experiment.MS medium without phytohormone was found as an appropriate basal one for growth of hairy roots.NAA, 6-BA, IBA added in medium, to some extent, had an inhibitive effect on growth of hairy roots.The optimal components of liquid medium for hairy roots growth were as follows:1.5% of sucrose, nitrogen source concentration same as MS medium, 3.75mmol/L phosphate, initial pH 6.1.100μmol/L of salicylic acid could increase secretion of resveratrol glucoside and improve production of resveratrol in hairy roots of peanut.
