Research progress of rolling circle amplification of DNA

  • 摘要: 作为一种等温DNA扩增技术,滚环扩增(Rolling Circle Amplification,RCA),以其简单和高效的特点得到了广泛的研究和应用。一般通过与靶序列进行杂交,特异性地生成环状DNA来达到DNA检测的目的。RCA已经广泛应用于免疫检测、原位检测、SNP检测等方面。本文将对RCA的工作原理、技术进展、应用前景以及存在的问题等进行介绍。 

     

    Abstract: As an isothermal DNA amplification technology, the well-known RCA (rolling circle amplification) has gained wide attention for its simplicity and high efficiency. Generally, forming specifically the circular DNA through hybridizing to the target molecule is the key point to realize specific detection. RCA has been extensively applied to immunoassay, in situ detection, SNP investigation and microarray assay. This review would introduce the principle, technological development, applicable prospect and the present problems of RCA.
