高效液相色谱法测定番茄及其制品中番茄甙的含量

Determination of tomatine in tomato and its processed products by high performance liquid chromatography

  • 摘要: 建立了固相萃取-高效液相色谱(HPLC)测定番茄及番茄制品中番茄甙的方法。样品中的番茄甙经过甲醇提取,PCX固相萃取法净化,乙酰化后,采用高效液相色谱法进行测定。以乙腈和水(90:10)为流动相,经过C18色谱柱分离,紫外检测器检测。结果表明:番茄甙的检出限为0.5mg/kg,方法回收率为90.5%~93.5%,相对标准差为1.7%~3.2%。该方法准确、灵敏、适用于番茄及番茄制品中番茄甙的测定。 

     

    Abstract: A method was developed for determination of tomatine in tomato and its processed products by high performance liquid chromatography (HPLC) .The tomatine in sample was extracted by methanol, cleaned up with PCX solid-phase extraction cartridge, then acetylated by acetic anhydride and determined by HPLC.The mobile phase consisted of 90% acetonitrile and 10% water.The tomatine was separated on C18 column and determined with UV detector at 205nm.The results showed:the detection limit of tomatine was 0.5mg/kg, the average recoveries were from 90.5% to 93.5%, and the relative standard deviations (RSDs) was from 1.7% to 3.2%.This method was accurate, sensitive and effective for determination of tomatine in tomato and its processed products.
