牛初乳生长因子粗提物对CaCO-2细胞增殖和迁移的影响

摘要: 本文探讨牛初乳生长因子粗提物对CaCO-2细胞生长和迁移的作用,为治疗人类胃肠炎症提供一种安全可靠的方法。给予CaCO-2细胞一定剂量的牛初乳生长因子粗提物,通过MTT法,分别测定12、24、36h细胞的增殖率;用BCA试剂测定CaCO-2总蛋白质含量;用划线法测定细胞的迁移能力。结果表明:与对照组相比,细胞增殖率与牛初乳生长因子粗体物有剂量依懒性。培养12h时,100ng/mL牛初乳生长因子即能显著刺激细胞增殖,达到16.3%(p<0.05),1μg/mL时,有最大的增殖率20.6%;培养24,36h时,1μg/mL牛初乳生长因子粗提物对细胞有最大增殖率,分别达到30.8%和40.0%,但是此时显著低于10%胎牛血清对细胞的增殖率;细胞蛋白质和迁移速度也与牛初乳生长因子粗提物浓度呈现剂量依懒性关系,在10μg/mL时,达到最大含量和迁移速度;本研究证实牛初乳生长因子粗提物能显著促进细胞增殖,蛋白质含量增加和细胞迁移速度。Abstract: In this study,the effect of colostrum growth factors extracts on the proliferation and migration of CaCO-2cells was investigated,which provided a safe and reliable way for the treatment of human gastrointestinal inflammation.Proliferation rate of CaCO-2 cells were determined by MTT assay at 12,24,36 h respectively,CaCO-2total protein content were determinated by BCA reagent. The migration of CaCO-2 cells were determined by scratch assay after given a dose of colostrum growth factors extracts. Results indicated that CaCO-2 cells were related with colostrum growth factors extracts in a way of dose-dependent manner,compared with the control group. Colostrum growth factors with 100 ng/m L could stimulated dramatically the proliferation of CaCO-2 cells at culturing 12 h,by 16.3%( p < 0.05). When the concentration was 1μg/m L,the cells had the maximum proliferation rate by 20.6%. Colostrum growth factors extracts with 1μg/m L could stimulate cells with maximum proliferation at culturing 24,36 h,increased by 30.8% and 40.0%,respectively,but significantly lower than the proliferation rate by the medium containing 10% fetal bovine serum. Cells protein content and migration speed also had related with colostrum growth factors extracts in a way of dose-dependent manner. There were the maximum protein content and migration speed with 10μg/m L.These results demonstrated that colostrum growth factors extracts significantly accelerated cells proliferation rate,increased cell protein content and migration speed.
