海洋胶原低聚肽的分离纯化及其抗氧化活性研究

摘要: 通过总抗氧化能力、羟基自由基清除能力、超氧阴离子自由基清除能力及还原能力实验,对海洋胶原低聚肽的体外抗氧化活性进行考察,然后采用反相高效液相色谱(RP-HPLC)对海洋胶原低聚肽进行分离纯化,测定收集得到的11个组分的总抗氧化能力。结果表明,海洋胶原低聚肽具有一定的总抗氧化能力、羟基自由基和超氧阴离子自由基清除能力,并且具有一定的还原能力;RP-HPLC分离纯化后,5个组分的抗氧化活性显著提高,对总肽整体的抗氧化活性起到了重要的作用,并且疏水性肽组分对抗氧化活性的贡献大于亲水性肽组分。Abstract: The antioxidant activities in vitro of marine collagen oligopeptides (MCOP) were evaluated from four respects by determination of the total antioxidant capacity and the scavenging activities against hydroxyl radical and superoxide radical as well as the reducing power.Then, MCOP was separated and purified by reversed phase high performance liquid chromatography (RP-HPLC) and the total antioxidant capacity of 11 fractions collected was assayed.Results indicated that MCOP exhibited total antioxidant capacity, scavenging activities against hydroxyl radical and superoxide radical and reducing power.After separation and purification by RP-HPLC, 5 fractions showed much higher total antioxidant capacity than MCOP, and played an important role in the total antioxidant activity of MCOP.Additionally, hydrophobic peptides in the MCOP presented higher activities than the hydrophilic ones.
