Effects of Anwulignan on the Hepatic Injury Induced by Intestinal Ischemia-Reperfusion in Rats

Abstract
Objective: To observe the effect and mechanism of Anwulignan on remote hepatic injury induced by intestinal ischemia-reperfusion (II/R) in rats.Methods: SD rats were randomly divided into sham operation (Sham) group, ischemia-reperfusion (II/R) group, sham operation + Anwulignan (Sham + Anwu) group, and II/R + Anwulignan (II/R Anwu) group.After sham operation and II/R operation, the abdominal aorta blood and the liver were taken. The pathological changes of liver tissue were observed under light microscope, serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activity, liver reactive oxygen species (ROS), protein kinase C (PKC) content, superoxide dismutase (SOD), catalase (CAT), and glutathione-S-transferase (GST) activity, as well as malondialdehyde (MDA) and 8-hydroxy-2'-deoxyguanosine (8-OHdG) content were detected. Results:Under the light microscope, the liver tissue structure of sham group was normal; The histopathological changes were observed in II / R group; Compared with II / R group, the injury in II / R + Anwu group was significantly reduced. Compared with Sham group, serum ALT and AST activities, liver ROS, MDA, 8-OHdG and PKC contents in II/R group increased extremely significantly (P<0.01), liver SOD and GST activities decreased extremely significantly (P<0.01), CAT activity decreased significantly (P<0.05). However, there were no significant changes in these indicators in Sham + Anwu group (P > 0.05). When compared with II/R group, the serum AST, liver MDA, 8-OHdG contents in the II/R + Anwu group decreased extremely significantly (P<0.01), the serum ALT, liver ROS and PKC level decreased significantly (P<0.05), the activities of liver SOD, CAT and GST increased extremely significantly (P<0.01). Conclusion: Anwulignan can improve the antioxidant capacity of liver tissues and ameliorate the liver injury induced by II/R rats.
