Study on In Vivo and In Vitro Antioxidant Activity of Collagen Oligopeptide Chelated Zinc from Puffer Skin

Abstract
Collagen oligopeptide chelated zinc was prepared from puffer skin. Its composition and structure were characterized. Taking 1,1-diphenyl-2-trinitrobenzene hydrazine free radical(DPPH·),hydroxyl free radical(·OH)scavenging ability,ferric iron(Fe3+)reducing power as indicators,the in vitro antioxidant capacity of collagen oligopeptide chelated zinc was evaluated. The experimental results showed that both collagen peptide and zinc chelate had DPPH free radical and hydroxyl free radicals scavenging activities,and ferric iron(Fe3+)reducing power. They also had good dose-effect relationship with the sample concentration. Collagen oligopeptide chelated zinc showed greater antioxidant ability than collagen oligopeptide. Collagen oligopeptide chelated zinc could significantly improve the total antioxidant capacity(T-AOC),glutathione(GSH)and total superoxide dismutase(T-SOD)activity in the serum of Zn-deficient rats,and obviously reduce the content of malondialdehyde(MDA)(P<0.05,0.01). Collagen oligopeptide chelated zinc from puffer skin has excellent antioxidant activity in vitro and in vivo,which is expected to be used as a new antioxidant in biological products and food industry.
