Optimization of Polysaccharides and Fibrinolytic Enzyme Co-production from Cordyceps militaris through Solid State Fermentation

Abstract
In the present work,the polysaccharides and the fibrinolytic enzyme were produced by solid-state fermentation of Cordyceps militaris,and corn flour was used as the main raw material. The medium and culture conditions were optimized by single factor experiment and orthogonal experiment using the polysaccharide production and activities of fibrinolytic enzyme as indicators. The results showed that the medium was composed of corn flour and bran at a ratio of 18:2 (g/g),the solid-liquid ratio of 1:1 (m/V),6%(V/m)of inoculation volume,fermentation temperature 23 ℃,and 3 days of the fermentation time. Under the optimized conditions,the yield of polysaccharide from the extract was 3.23 mg/mL and the degradation zone area of Cordyceps militaris product reached 169.34 mm2 in the fibrin plate. These results could provide a basis to produce functional foods of Cordyceps militaris.
