联合使用羧甲基壳聚糖与阿霉素抗肿瘤效应
Effects on tumor cells by combination with carboxymethyl-chitosan and adriamycin

摘要: 为了研究羧甲基壳聚糖(Carboxymethyl-chitosan,CM-CTS)与阿霉素(Adriamycin,ADM)的联合抗肿瘤作用。通过细胞平板克隆实验、细胞划痕实验来评价羧甲基壳聚糖和阿霉素对Hela细胞增殖和迁移能力的影响,B16实体瘤模型用于评价体内抗肿瘤活性。结果表明,CM-CTS与ADM联合使用对Hela细胞克隆形成的抑制率为72.65%,极显著高于单独使用CM-CTS(31.47%)或ADM(44.09%)(p<0.01)。CM-CTS与ADM联合使用对Hela细胞迁移的抑制率为84.48%,极显著高于CM-CTS(37.47%)或ADM(56.29%)(p<0.01)单独用药时的抑制作用。体内动物实验结果显示,CM-CTS和ADM单独使用时,在活的有机体内对肿瘤细胞的生长抑制率分别为27.68%、16.96%,当两种药物联合使用时,对肿瘤生长的抑制率为47.32%,具有协同效应。结果说明,联合用药可以显著抑制宫颈癌细胞Hela的增殖和迁移。Abstract: The effect of anti-tumor combining carboxymethyl-chitosan(CM-CTS)with adriamycin(ADM)was studied. Colony forming test and trauma test were used to evaluate the effects of CM-CTS and ADM on cell proliferation and cell migration of Hela in vitro. Animal experiment was used to evaluate the anti-tumor activity of CM-CTS and ADM in vivo. The results showed that combining CM-CTS with ADM could inhibit clone forming rate of Hela by 72.65%,which was significantly higher than treating Hela with CM-CTS(31.47%)or ADM(44.09%)only(p<0.01). The migration ability of Hela could be inhibited when Hela was treated by CM-CTS and ADM together. And the inhibition rate was 84.48%,which was significantly higher than that of CTS(37.47%)or ADM(56.29%)(p<0.01). Animal experiment showed that the suppression rate of CM-CTS or ADM was 16.96% or 27.68%(p<0.01). While two-drug combination had a synergistic effect,which inhibited the tumor growth by 47.32%. Therefore,synergistic effect of CM-CTS and ADM could effectively inhibit cell proliferation and migration of Hela.
