紫苏叶体外抗氧化和降糖活性分析

Antioxidant and Hypoglycemic Activities of Perilla Leaves in Vitro

  • 摘要: 为了研究紫苏叶及其提取物的体外抗氧化和降糖活性,本研究利用快速粘度分析仪制备紫苏叶基淀粉糊样品并进行体外模拟胃肠消化,采用福林酚法测定消化前后总酚含量,并通过研究消化前后样品对DPPH和ABTS+自由基的清除能力分析其抗氧化性能,用3,5-二硝基水杨酸(DNS)法测定紫苏叶及其提取物对大米淀粉消化过程中还原糖释放量的影响。结果表明,随着活性物质添加量从5%增加到15%,消化过程中还原糖释放曲线线下所围面积(Area under the Curve,AUC)值总体呈下降趋势,其中添加15%一次纯化物的样品AUC值(30.86 mg葡萄糖/g)最低;经模拟胃肠消化后总酚含量均有显著增加,添加15%一次纯化物的样品多酚含量(81.04 mg GAE/100 g DW)最高,且消化后清除DPPH自由基和ABTS+自由基的能力显著(P<0.05)提高,其中添加10%一次纯化物对DPPH自由基的清除能力(38.69%)最高,添加15%一次纯化物对ABTS+自由基的清除能力(57.25%)最高。该研究表明紫苏叶及其提取物可以抑制大米淀粉体外升糖反应,且具有较好的抗氧化能力。

     

    Abstract: In order to study the antioxidant and hypoglycemic activities in vitro of Perilla leaves and their extracts, the starch paste samples with Perilla leaves were prepared using a rapid viscosity analyzer, and then in vitro simulated gastrointestinal digestion was conducted. The total phenolic content before and after digestion was measured by the Folin phenol method. The antioxidant properties of the samples were analyzed by their ability to scavenge DPPH and ABTS+ free radicals before and after digestion. The effects of Perilla leaves and their extracts on the release of reducing sugars during the digestion of rice starch were determined by 3,5-dinitrosalicylic acid. The results showed that as the amount of active substance added increased from 5% to 15%, the area under the curve (AUC) of reducing sugar released during the digestion process showed an overall downward trend, the sample with 15% primary purified powder had the lowest AUC value (30.86 mg glucose/g). After simulated gastrointestinal digestion, the total phenol content significantly increased, the sample with 15% primary purified powder had the highest content of total polyphenol (81.04 mg GAE/100 g DW). The ability of scavenging DPPH and ABTS+ free radical was improved significantly (P<0.05). The sample with 10% primary purified powder had the highest scavenging ability (38.69%) of DPPH radicals, and the sample with 15% primary purified powder had the highest scavenging ability (57.25%) of ABTS+ radicals. It was indicated that Perilla leaves and their extracts could inhibit the glycemic reaction of rice starch in vitro and had strong antioxidant capacity.
