Analysis on the Difference of Mineral Element Content in Yak Meat

摘要: 目的:分析西藏昌都市类乌齐县牦牛肉中矿质元素的含量差异性。方法:本研究以西藏昌都市类乌齐县(卡玛多乡、类乌齐镇、伊日乡、长毛岭乡、岗色乡、桑多镇、宾达乡、尚卡乡、甲桑卡乡、吉多乡)431份牦牛肉为研究对象。采用微波消解-电感耦合等离子体质谱法测定牦牛肉中的K、Na、Ca、Mg、Se、Sr、Mn、Zn、Fe等9种矿质元素含量。分析不同地域、不同部位牦牛肉中矿质元素含量的差异性。结果:类乌齐牦牛肉不同部位矿质元素存在极显著差异(P<0.01);类乌齐牦牛肉臀部肉中Ca、Se元素存在极显著差异(P<0.01),Zn、Fe元素存在显著差异(P<0.05);类乌齐牦牛肉前腿肉中K、Na、Ca、Mg、Se、Mn元素存在显著差异(P<0.05);类乌齐牦牛肉后腿肉中K、Mg、Se、Zn元素存在极显著差异(P<0.01),Na、Ca存在显著差异(P<0.05);类乌齐牦牛肉里脊肉中K、Na、Mg元素存在极显著差异(P<0.01),Ca、Mn、Zn元素存在显著差异(P<0.05);类乌齐牦牛肉腱子肉中K、Mn存在显著差异(P<0.05),Sr、Zn、Fe元素存在极显著差异(P<0.01);类乌齐牦牛肉牛腩肉中Ca、Fe元素存在显著差异(P<0.05)。结论:不同地区的牦牛肉K、Na、Mg含量均较高,Ca、Zn、Fe次之,Se、Sr、Mn含量较低。不同部位牦牛肉矿质元素中里脊肉K、Na、Mg,牛腩Ca,后腿Se,臀肉Sr、Mn,腱子Zn、Fe具备含量和品质优势。Abstract: Objective: Analyze the differences in mineral element content in yak meat from Leiwuqi County, Changdu City, Tibet. Methods: This study focused on 431 yak meat samples from Leiwuqi County, Xizangchang City, including (Kamado Township, Leiwuqi Township, Yiri Township, Changmaoling Township, Gangse Township, Sangdo Township, Binda Township, Shangka Township, Jiasangka Township, Jiduo Township). The contents of K, Na, Ca, Mg, Se, Sr, Mn, Zn and Fe in yak meat were determined by microwave digestion ICP-MS. The difference of mineral element content in yak meat from different regions and parts were analyzed. Results: There were extremely significant differences in mineral elements in different parts of the meat of similar Uqi yaks (P<0.01). Extremely significant differences in Ca and Se elements (P<0.01) and significant differences in Zn and Fe elements (P<0.05). There were significant differences in K, Na, Ca, Mg, Se, and Mn elements (P<0.05), extremely significant differences in K, Mg, Se, Zn elements (P<0.01), and significant differences in Na and Ca (P<0.05). There were extremely significant differences in K, Na, and Mg elements (P<0.01), significant differences in Ca, Mn, and Zn elements (P<0.05). There were significant differences in K and Mn (P<0.05), extremely significant differences in Sr, Zn, and Fe elements (P<0.01), and significant difference in Ca and Fe elements (P<0.05). Conclusion: Yak meat from different regions had higher levels of K, Na, and Mg,Ca, Zn, and Fe take second place, the content of Se, Sr, and Mn was relatively low. The mineral elements in different parts of yak meat included K, Na, Mg in tenderloin, Ca in brisket, Se in hind legs, Sr, Mn in buttocks, and Zn, Fe in tendons, which had advantages in content and quality.

牦牛(Yak)长年生活在海拔3000 m以上的高寒地区[1],适应高寒生态条件,耐粗饲、耐劳,善走陡坡险路、雪山沼泽。牦牛完全依赖于高山天然草场,使得牦牛肉成为优良的肉品来源[2−4]。牦牛肉富含蛋白质和Ca、P等微量元素[5],脂肪含量极低[6],对增强人体免疫力、细胞活力和器官功能均有显著作用[7]。
西藏地方类群牦牛种类较多,不同地域牦牛肉的品质和口感存在很大的差异[8],不同地域环境中矿物元素含量存在差异,这使得植物中矿质元素含量存在差异,由此以植物为饲料的动物体内矿质元素含量也存在差异。目前,对地方优势品种类乌齐牦牛各部位的矿质元素数据几乎没有。类乌齐牦牛产区地势西北高,东南低,整个地貌景观呈山原类型,造就山间河谷深切、沟壑纵横、峡谷众多、坡向分明的特点[9]。类乌齐牦牛肉为中国国家地理标志产品,类乌齐牦牛放养于海拔3700 m以上的类乌齐县天然牧场中,“天然、绿色、原生态”的类乌齐牦牛以膘肥体壮、肉质颜色鲜红、致密有弹性、口味浓郁,营养价值高而享誉区内外[10]。目前,对于牦牛肉品质的研究主要集中在不同饲养模式[11−14]、不同品种牦牛肉营养品质评价与对比分析[15−17],不同部位肉对其加工品质的影响[18−20]。但是,对不同部位矿物质成分含量差异性分析报道不多[21],针对类乌齐牦牛肉中矿物质元素的含量差异性分析的更是少之又少。
本研究以类乌齐10个不同地区的6个不同部位431个牦牛肉样本为实验材料,分析比较不同地区、部位牦牛肉中的矿物元素含量及差异性,通过比较分析得出类乌齐牦牛肉不同地区、部位矿质元素的含量优势,明确不同地区、部位牦牛肉的品质优势,进一步以数据为支撑保护地理标志产品,为合理开发利用类乌齐牦牛肉、提高经济效益提供科学依据。
1. 材料与方法
1.1 材料与仪器
牦牛肉 西藏昌都市类乌齐县(卡玛多乡、类乌齐镇、伊日乡、长毛岭乡、岗色乡、桑多镇、宾达乡、尚卡乡、甲桑卡乡、吉多乡)屠宰场。将各乡镇每年需要宰杀的牦牛集中宰杀,样品于现场采集,总共431份(见表1),牦牛肉样品放于冷藏箱(0~4 ℃)内带回实验室,于−20 ℃保存待测;Fe、Mn、Cu、Zn、K、Ca、Mg、Na、Se、Sr的标准溶液1000 μg/mL 国家有色金属及电子材料分析测试中心;硝酸 优级纯,国药集团化学试剂有限公司;氩气 纯度≥99.999%,成都龙泰工业气体有限公司。

1.2 实验方法
1.2.1 样品处理
样品处理、元素的测定参考GB 5009.268-2016《食品安全国家标准 食品中多元素的测定》[22]。
1.2.2 检测条件
ICP-MS仪器工作参数为频射功率1550 W,ICP-MS 工作条件参数为射频功率1300 W,等离子体气流量15 L/min,载气流量1.0 L/min,测定模式KED/STD,样品提升速率3 r/s,采样深度9.0 mm,进样冲洗时间20 s,扫描次数30次,重复次数3次,蠕动泵提升转速48 r/min。
1.3 数据处理
试验数据利用Excel进行数据统计,应用SPSS17.0统计软件进行方差分析,P<0.05表示差异显著,P<0.01表示差异极显著,使用Origin进行绘图。
2. 结果与分析
2.1 不同部位牦牛肉矿质元素含量差异性比较
比较不同部位的牦牛肉矿质元素品质优势。从表2 中可知,类乌齐牦牛肉不同部位K元素:里脊与其它五部位差异极显著(P<0.01),腱子和牛腩与臀肉、前腿、后腿之间存在极显著差异(P<0.01);Na元素:里脊、腱子与其它五部位间差异均极显著(P<0.01);Ca元素:腱子、牛腩与其它四部位差异极显著(P<0.01),里脊、前腿与腱子、牛腩之间存在极显著差异(P<0.01);Mg元素:牛腩、腱子与其它五部位差异显著(P<0.01);Se元素:臀肉、后腿与其它四部位间差异极显著(P<0.01);Sr元素:牛腩与其它五部位间差异显著(P<0.01),臀肉与前腿、里脊之间差异极显著(P<0.01);Mn元素:前腿、牛腩与其它四部位间差异显著(P<0.01);Zn元素:臀肉、后腿与其它四部位差异极显著(P<0.01);Fe元素:臀肉、前腿、后腿与其它三部位差异极显著(P<0.01)。K元素:里脊肉含量最高为4059.630 mg/kg;Na元素:里脊肉含量最高为679.521 mg/kg;Ca元素:牛腩含量最高为117.681 mg/kg;Mg元素:里脊含量最高为289.360 mg/kg;Se元素:后腿含量最高为1.117 mg/kg;Sr元素:臀肉含量最高为0.476 mg/kg;Mn元素:臀肉含量最高为0.451 mg/kg;Zn元素:腱子含量最高为62.961 mg/kg;Fe元素:腱子含量最高为58.481 mg/kg。

比较不同乡镇臀部肉中矿质元素的含量优势,由表3可知,类乌齐牦牛肉臀部肉中K、Na、Mg、Sr、Mn含量在十个乡镇之间均没有差异(P>0.05)。Ca元素:长毛岭乡、宾达乡之间差异极显著(P<0.01);Se元素:吉多乡与除甲桑卡乡之外的八乡镇间差异极显著(P<0.01);Zn元素:长毛岭乡、吉多乡之间差异显著(P<0.05);Fe元素:尚卡乡与桑多镇、宾达乡存在显著差异(P<0.05)。K元素:吉多乡含量最高为3640.410 mg/kg;Na元素:岗色乡含量最高为477.701 mg/kg;Ca元素:长毛岭乡含量最高为47.543 mg/kg;Mg元素:长毛岭乡含量最高为289.360 mg/kg;Se元素:吉多乡含量最高为2.649 mg/kg;Sr元素:伊日乡含量最高为0.539 mg/kg;Mn元素:类乌齐镇含量最高为0.528 mg/kg;Zn元素:长毛岭乡含量最高为52.774 mg/kg;Fe元素:尚卡乡含量最高为53.566 mg/kg。

研究显示K、Na在体内维持细胞的内外渗透平衡以及神经细胞膜的生物兴奋性等方面具有重要作用[23];Se是动物体内必需的微量元素,是体内谷胱甘肽过氧化酶重要组成部分[21],Fe缺乏会导致贫血[24];Cu是体内蛋白质及酶的重要组成,参与许多酶的反应,催化血红蛋白的合成[25];Ca是肌肉收缩和肌原纤维降解酶系的激活剂,能够显著改善牛肉的嫩度[26−27];Mg作为重要的辅酶,参与多种代谢反应,能够通过抑制屠宰前应激所导致的糖原分解反应来改善肉质,且镁含量高低可对肌肉的pH产生影响[28];Zn是脑细胞生长的关键,促进人体的生长发育、伤口和创伤的愈合,影响维生素A的代谢和正常视觉,维持男性正常的生精功能[29],人体中Zn主要来源于食物,特别是红肉如牛肉等;Mn可减少血管内血栓的形成, 降低动脉硬化的危险[30];Sr能够促进骨骼吸收、预防骨质疏松症,对非酒精性脂肪肝的预防方面都有着积极的作用[31]。类乌齐牦牛肉中富含矿质元素,可作为人们日常生活中一种非常好的补充矿质元素营养的食品,对人体的健康非常有益处。
不同部位牦牛肉矿质元素含量差异性比较结果显示,类乌齐牦牛肉不同部位矿质元素存在极显著差异(P<0.01);不同乡镇臀部肉矿质元素含量差异性比较结果显示,类乌齐牦牛肉臀部肉中K、Na、Mg、Sr、Mn含量在十个乡镇之间均没有差异,Ca、Se元素存在极显著差异(P<0.01),Zn、Fe元素存在显著差异(P<0.05);不同乡镇前腿肉矿质元素含量差异性比较结果显示,类乌齐牦牛肉前腿肉中Sr、Zn、Fe含量在十个乡镇之间均没有差异,K、Na、Ca、Mg、Se、Mn元素存在显著差异(P<0.05);不同乡镇后腿肉矿质元素含量差异性比较结果显示,类乌齐牦牛肉后腿肉中Sr、Mn、Fe含量在十个乡镇之间均没有差异,K、Mg、Se、Zn元素存在极显著差异(P<0.01),Na、Ca存在显著差异(P<0.05);不同乡镇里脊肉矿质元素含量差异性比较结果显示,类乌齐牦牛肉里脊肉中Se、Sr、Fe含量在十个乡镇之间均没有差异,K、Na、Mg元素存在极显著差异(P<0.01),Ca、Mn、Zn元素存在显著差异(P<0.05);不同乡镇腱子肉矿质元素含量差异性比较结果显示,类乌齐牦牛肉腱子肉中Na、Ca、Mg、Se含量在十个乡镇之间均没有差异,K、Mn存在显著差异(P<0.05),Sr、Zn、Fe元素存在极显著差异(P<0.01);不同乡镇牛腩肉矿质元素含量差异性比较结果显示,类乌齐牦牛肉牛腩肉中K、Na、Mg、Se、Sr、Mn、Zn含量在五个乡镇之间均没有差异,Ca、Fe元素存在显著差异(P<0.05)。牦牛肉中矿物质元素含量的差异,由多方面的因素综合作用而产生。不同年龄段牦牛、不同品种牦牛、不同部位牦牛肉以及不同地区的土壤、水源、气候等因素都会直接或间接影响牦牛肉矿物质元素的累积,最终造成矿质元素在不同地区和不同部位间的差异性。
通过对西藏类乌齐县不同乡镇牦牛肉不同部位中9种矿质元素的含量测定及分析,可知,不同地区的牦牛肉K、Na、Mg含量均较高,Ca、Zn、Fe次之,Se、Sr、Mn含量较低。其中类乌齐县牦牛肉中含量最高的常量元素和微量元素分别是K和Zn。不同部位牦牛肉矿质元素含量:里脊肉K、Na、Mg元素含量最高,牛腩Ca元素含量最高,后腿Se元素含量最高为1.117 mg/kg,臀肉Sr、Mn元素含量最高,腱子Zn、Fe元素含量最高。研究结果表明, 431份类乌齐牦牛肉中矿质元素K、Na、Mg、Ca、Zn、Fe、Se、Sr、Mn的平均值分别为2840.373、490.037、258.291、57.214、48.955、43.167、0.692、0.389、0.374 mg/kg。10乡镇不同部位矿质元素含量差异性比较显示,每个部位在各个乡镇之间均有不同程度的差异。不同乡镇臀部肉矿质元素含量:吉多乡K、Se元素含量最高,岗色乡Na元素含量最高,长毛岭乡Ca、Mg、Zn元素含量最高,伊日乡Sr元素含量最高,类乌齐镇Mn元素含量最高,尚卡乡Fe元素含量最高。不同乡镇前腿肉矿质元素含量:尚卡乡K、Zn元素含量最高,宾达乡Na元素含量最高,吉多乡Ca、Mg、Se、Fe元素含量最高,卡玛多乡Sr元素含量最高,长毛岭乡Mn元素含量最高。不同乡镇后腿肉矿质元素含量:类乌齐镇K、Na、Mg、Se元素含量最高,长毛岭乡Ca元素含量最高,岗色乡Sr元素含量最高,伊日乡Mn元素含量最高,甲桑卡乡Zn、Fe元素含量最高。不同乡镇里脊肉矿质元素含量:岗色乡K、Na元素含量最高,长毛岭乡Ca元素含量最高,甲桑卡乡Mg、Fe元素含量最高,吉多乡Se元素含量最高,尚卡乡Sr元素含量最高,宾达乡Mn元素含量最高,类乌齐镇Zn元素含量最高。不同乡镇腱子肉矿质元素含量:桑多镇K、Na元素含量最高,甲桑卡乡Ca、Zn元素含量最高,岗色乡Mg、Se、Mn、Fe元素含量最高,吉多乡Sr元素含量最高。不同乡镇牛腩肉矿质元素含量:岗色乡K元素含量最高,尚卡乡Na、Se、Mn、Zn元素含量最高,宾达乡Ca、Sr元素含量最高,桑多镇Mg元素含量最高,吉多乡Fe元素含量最高。
由于不同地区牦牛肉不同部位的肌肉生理功能、组织结构存在差异, 矿物元素在不同部位肌肉中的分布与沉积存在很大差异[32−35],检测结果可能受检验人员、样品制备、检验方法、实验环境、仪器、材料等因素影响,因此会存在一定的结果差异。本研究明确了类乌齐县牦牛肉不同地域和不同部位中9种矿质元素的含量及差异,能够有针对性的为生产提供数据指导,做到目的明确的生产和消费。为西藏类乌齐牦牛产业的精准长远发展提供数据支撑和理论依据。由于西藏牦牛肉地方种类丰富,在精准分割方面还存在不足,很难全面精准提供样品研究,致使研究不全面。今后应加大对西藏地方不同品种、不同部位牦牛肉的深入研究,为西藏牦牛产业的精准长远发展提供数据支撑和理论依据。
