Abstract:
Objective:To analyze the components of coix seed oil in Guizhou and study the effects of different concentrations of coix seed oil on osteoclast activity. Methods:The ethanol-assisted extraction of coix seed oil in Guizhou was carried out by anhydrous ethanol and the gas chromatography-mass spectrometry (GC-MS) analysis of fatty acid residues was carried out. Estradiol (10
-8 mol/L) was used as a positive control. Rabbit bone marrow cells were induced with 1
α, 25-(OH)
2Vit D
3 to obtain osteoclasts. The effects of coix seed oil in Guizhou on osteoclast activity were analyzed by TRAP+cell number, TRAP activity and bone sag area. Results:The fatty acid residues of hazelnut oil produced by GC-MS were mainly myristic acid, palmitic acid, octadecanoic acid (stearic acid), and eighteen carbon monoenoic acid (oleic acid), octadecadienoic acid (linoleic acid), behenic acid, squalene and
γ-sitosterol;further experiments showed that the concentration of coix seed oil in Guizhou (0.98, 1.95, 3.91, 7.81, 15.63, 31.25 μg/mL) had a certain inhibitory effect on osteoclast activity, 31.25 μg/mL in the high concentration group inhibited the activity of osteoclasts most significantly (
p<0.05), and showed a good concentration dependence.Conclusion:Coix seed oil in Guizhou has an inhibitory effect on osteoclast activity.
