不同壳聚糖脱乙酰度测定方法的适宜性研究

Suitability for Determination of Deacetylation Degree of Chitosan

  • 摘要: 为选择简便精准的壳聚糖脱乙酰度测定方法,从测定方法的准确度、精密度、通用性方面比较了目前常用的六种方法。结果表明,改良后的酸碱滴定法操作简单,准确度为97.01%,精密度的RSD为0.33%;线性电位滴定法在滴定时取点少,较双突跃滴定方便省时;紫外光谱法与红外光谱法均需特殊仪器;胶体滴定法测定变色灵敏,但对滴定速度要求严格,精密度较低。对改良后的酸碱滴定法、线性电位滴定法、紫外光谱法的通用性考察可知,上述三种方法通用性均较好,分子量低(≤ 100000)的壳聚糖脱乙酰度测定采用紫外光谱法,高粘度(≥ 400 mPa·s)壳聚糖脱乙酰度测定采用线性电位滴定法,其余类型壳聚糖脱乙酰度测定均可使用改良后的酸碱滴定法,操作简便,结果准确。

     

    Abstract: To choose a simple and accurate method for the determination of deacetylation degree of chitosan,the study evaluated six current methods from different aspects including the measurement accuracy,precision and versatility. The results showed that the modified acid-base titration was simple,and the accuracy was 97.01%,and the RSD of precision was 0.33%. Linear potentiometric titration needed less sample point,so it was more convenient and time-saving compared with double-dip titration. UV Spectroscopy and infrared spectroscopy required special equipment. Colloidal titration was sensitive to the change of color,but titration speed was limited and the precision was low. The versatility of modified acid-base titration,linear potentiometric titration and UV spectroscopy showed that they had good universality. UV Spectroscopy was adapted to determination of degree of deacetylation of chitosan with low molecular weight(≤ 100000). Linear potentiometric titration was adapted to chitosan with high viscosity(≥ 400 mPa·s). The degree of deacetylation of other kinds of chitosan could be determined by the improved acid-base titration method,which was simple and accurate.
