Physicochemical characteristic and antioxidation activity in vitro of water soluble polysaccharides from Zingiber mioga Rosc

摘要: 以蘘荷花轴为原料,通过水提醇沉法提取制备蘘荷水溶性多糖(water-soluble polysaccharides from Zingiber mioga Rosc.,ZMPs-W),采用化学和仪器分析方法研究了多糖的总糖含量、单糖组成、官能团特征、空间构像等理化性质,并通过体外抗氧化实验探讨了蘘荷水溶性多糖的抗氧化活性。研究结果表明:ZMPs-W是以半乳糖、葡萄糖、半乳糖醛酸、葡糖糖醛酸、阿拉伯糖、木糖和鼠李糖(各组分之间的摩尔百分比为:33.12∶18.79∶15.93∶11.47∶9.81∶5.98∶5.90)为主要组成成分的,含有羧基、硫酸基和吡喃环的多糖类物质,且具有三螺旋结构。其总糖含量和蛋白质含量分别为73.76%和0.35%;体外抗氧化实验表明:ZMPs-W具有一定的抗氧化活性,在本实验条件下,其清除羟基自由基、DPPH自由基以及螯合金属离子的能力随着添加剂量的增加而增强,其中清除羟基自由基和DPPH自由基的能力表现突出。Abstract: Water- soluble polysaccharides( ZMPs- W),extracted from floral axis of Zingiber mioga Rosc.by water extractingalcohol precipitation method were studied by the means of chemical and apparatus analysis.The antioxidation activities in vitro were also studied in this paper. The results showed that ZMPs- W were mainly constituted by galactose,glucose,galacturonic acid,glucuronic acid,arabinose,xylose,rhamnose( the mol ratio was 33.12 ∶ 18.79 ∶ 15.93 ∶ 11.47 ∶ 9.81 ∶ 5.98 ∶ 5.90). Triple helix structure,carboxyl group,sulfate group and pyran ring were found in the polysaccharides. The total sugar and protein content was 73.76% and 0.35%,respectively.Antioxidant experiment in vitro suggested that ZMPs- W had antioxidant activities.Under the experimental conditions,the antioxidant activities(·OH clearance,DPPH·clearance and Fe2 +chelation) increased with the increasing concentration.Especially,the clearance ablities of ·OH and DPPH· clearance were outstanding.
