Optimization of extraction conditions of Chinese chestnut peptides by response surface methodology and its DPPH· scavenging abilities

  • 摘要: 采用碱性蛋白酶酶解板栗蛋白制备抗氧化肽并测定其清除DPPH·能力。以水解度及DPPH·清除率为指标通过单因素实验初步得到板栗多肽提取工艺,再根据Box-Behnken中心组合实验设计原理,采用四因素三水平响应面分析法,建立了以加酶量、酶解温度、酶解p H、酶解时间与响应值DPPH·清除能力间的回归方程预测模型,确定了板栗抗氧化肽的最佳制备工艺条件:加酶量2.5%,酶解温度50℃,酶解p H9.5,酶解时间3.7 h。在此条件下DPPH·清除率为92.86%,与理论值基本相符。 
    Abstract: Antioxidant peptide was prepared by alkaline protease enzymolysising Chinese chestnut protein and its antioxidant activity determined.The craft was obtained with the degree of hydrolysis and the DPPH· scavenging ratio as indicators by single factor tests primarily,then according to Box- Behnken central composition design principle,the method of response surface analysis with 4 factors and 3 levels combined with Design Expert 8.0-statistical data analysis software was adopted to establish the regression equation prediction model of enzyme dose,enzymolysis temperature,enzymolysis p H and enzymolysis time and the DPPH· scavenging ratio as the response value.The optimum preparation conditions were established as follows: enzymedose 2.5%,enzymolysis temperature 50 ℃,enzymolysis p H9.5 and enzymolysis time 3.7 h.Under these conditions,the DPPH· scavenging ratio reached 92.86%,consistent with the theoretical value.
