玛咖总生物碱检测方法的建立及提取优化

Establishment of content determination and optimization extraction condition of total alkaloids in Maca

  • 摘要: 建立玛咖总生物碱含量的测定方法,对不同产地的玛咖总生物碱含量进行比较研究。以氧化苦参碱作为对照品,筛选酸性染料比色法条件,并利用响应面分析法优化样品处理方法,建立测定方法。结果酸性染料比色法的条件为缓冲液(p H=3.5)加入量3 m L,溴甲酚绿加入量3 m L,测定波长为412 nm;最佳样品处理条件为液料比23∶1(m L/g)、提取温度82℃、提取时间2.1 h。此条件下,云南黄色玛咖的总生物碱含量为1.3 mg/g,在五种玛咖中,西藏黑玛咖的总生物碱含量最高为8.4 mg/g。此法操作简便、精密度高、稳定性好、具良好的可重复性,可用于玛咖中总生物碱的含量测定。 

     

    Abstract: To establish a method for the assay of the total alkaloids in Maca,and investigate the contents from different origins. With the oxymatrine as reference,the method was established by purified acid dye colorimetry and optmized the extraction condition by response surface method. Acid dye colorimetric conditions was as follows : adding p H3. 5 buffer 3 m L and bromocresol green colour reagent 3 m L, then detemining at the wavelength 412 nm. The optimal extraction conditions was found to be ratio of liquid to solid 23 ∶1(m L/g),temperature 82 ℃,time 2.1 h. The extraction efficiency of total alkaloids from Yunnan yellow Maca under these conditions was 1.3 mg/g compared to 8.4 mg/g from Tibet black Maca,highest among the five species investigated. This method was simple,highly sensitive,accurate and reproducible,which could be used for determination of the total alkaloids in Maca.
