Abstract:
The purpose of this work was to study the distribution of Se in Xinchun No.36 triticale in Xingjiang province. Through single factor experiment and response surface method, a systematic inspection of selenoproteins,selenium polysaccharide,selenium nucleic acid and selenoproteins of solubility in different solvents was made. The results showed that content of selenoproteins,selenium ploysaccharide and nucleic acid selenium was(240.52±0.3),(32.45±0.21) and(4.65±0.05) mg/g respectively. The content of selenoproteins of alkali-soluble,water-soluble,salt-soluble and alcohol-soluble was(146.21±0.3),(41.52±0.6),(19.40±0.06)and(13.65±0.12) mg/g respectively. Furthermore,in order to get more alkali-soluble selenoproteins,the optimal extraction technology was obtained by the single factor test and the response surface design experiments. The optimal extraction conditions were Na OH 0.17 mol/L,acidification at p H4.7,saturation of(NH4)2SO466.0%,the ratio of solid to liquid 1 ∶10, the predicted rate of protein extraction was 76.5593%, but the actual value of experiment reached 76.7%.
