Abstract:
Response surface analysis (RSA) was used to optimize process conditions of agarose extraction. The main factors discussed were:DEAE-cellulose content, extraction temperature and extraction time on influencing in gel strength and sulfate radical content. Using the response surface method to optimize the extraction process, the best parameters were the quantity of DEAE-cellulose 55 g, extraction temperature 80℃ and extracting time 2.5h. With the optimized parameters, the sulfate radical content and gel strength of agarose extracted were 0.17% and 1210g/cm2, respectively. IR spectrum analysis showed that the agrose was similar to standard agarose. The genomic DNA electrophoresis proved that it had a good electrophoretic performance.
