Optimization of ultrasound assisted extraction and content determination of polysaccharide from Cynomorium songaricum Rupr.

摘要: 目的:筛选优化得到锁阳多糖最佳超声提取工艺,研究锁阳多糖含量测定方法,测定比较锁阳样品中多糖含量情况。方法:以多糖得率为考察指标,苯酚-硫酸法为含量测定方法,采用单因素和(L9(34))正交实验考察锁阳多糖超声提取工艺,并在含量测定方法学研究的基础上测定比较各样品多糖含量。结果:锁阳多糖超声提取最佳工艺为:超声功率420W,料液比1∶35,提取温度80℃,超声提取时间60min,提取2次。在此条件下,锁阳多糖平均得率可达4.51%(n=5,RSD=1.98%)。瓜州锁阳多糖含量明显较新疆和内蒙高,且瓜州三九锁阳多糖含量最高。结论:实验得到的超声提取工艺能明显提高锁阳多糖得率,实验采用的含量测定方法快速、简便,结果可靠,研究结果可为锁阳资源的合理开发及其产品的质量控制提供科学的参考依据。Abstract: Objective: To optimize ultrasonic extraction process of polysaccharide from Cynomorium songaricum Rupr.and determine the polysaccharide content in different samples.Msethod: The polysaccharide content was detected by phenol-sulfuric acid method.Use polysaccharide extraction yield as investigation index and extraction process was optimized by single factor and (L 9 (34) ) orthogonal experiment.Results: The optimum ultrasonic extraction conditions were: ultrasonic power (420W) , solid-liquid ratio (1 ∶ 35) , extraction temperature (80℃) , extraction time (60min) , and extraction times (2 times) .Under this condition, the extraction rate of polysaccharides reached 4.51% (n = 5, RSD = 1.98%) .The polysaccharide content of Guazhou samples was significantly higher than Xinjiang and Inner Mongolia;and polysaccharide content of Guazhou three-nine Cynomorium songaricum Rupr.was the highest in all samples.Conclusion: This ultrasonic extraction process could significantly improve the polysaccharides extraction rate.This method was rapid and simple;the experimental results were accurate and reliable.This study could provide reference data for rational development and quality control of Cynomorium songaricum Rupr and its products.
