羧甲基壳聚糖包覆薏苡仁油脂质体包覆率测定方法的研究

摘要: 建立了羧甲基壳聚糖对脂质体包覆率的测定方法。采用离心法将未包覆脂质体与包覆脂质体分离后,乙醇-超声法对未包覆脂质体进行破壁处理,磷脂经氯仿提取后,用硫氰铁铵法测定未包覆脂质体中磷脂的含量,根据总磷脂用量,计算羧甲基壳聚糖对脂质体的包覆率。在10000r/min条件下离心15min能实现较好的分离效果;当乙醇用量0.8mL超声处理15min时能充分打开未包覆脂质体;470nm为磷脂测定波长,磷脂在0.01~0.1mg/mL的浓度范围内,线性关系良好,经回收率实验、精密度及重现性实验证实该方法可行。Abstract: Ammonium thiocyanate method was applied to determine coating efficiency for carboxymethyl chitosan coated liposomes. The result showed that the wavelength for determination of phospholipid was at 470nm . At the concentration range of 0 . 01 ~ 0 . 1mg / mL , the content of phospholipid showed good linear relationship with the absorbance. Before determination, the uncoated liposomes were isolated from the coated ones by centrifugation operated at 10000r/min for 15min and broken by alcohol-ultrasound method with 0.8mL of ethanol and ultrasonication for 15min. Through recovery, precision, and reproducible experiments proved that the method was feasible.
