黄山贡菊抗氧化活性成分提取工艺优化与HPLC-MS分析

摘要: 探讨黄山贡菊的抗氧化活性化学成分。以FRAP及DPPH法分析抗氧化活性,采用响应面法优化黄山贡菊抗氧化活性成分的提取条件。在乙醇浓度60%,液料比35:1 (mL/g),超声时间25 min的条件下,黄山贡菊提取物的总抗氧化能力为(2.134±0.011) mmol/L(RSD=1.97%,n=3),与预测值(2.010 mmol/L)吻合很好。总黄酮当量为7.88 mg芦丁/L的黄山贡菊提取物的抗氧化能力相当于2.87 mmol/L亚铁离子的还原力;总黄酮当量为25.85 mg芦丁/L的提取物的DPPH自由基清除率为94.81%。采用HPLC-MS分析黄山贡菊提取物的抗氧化活性成分,共鉴定出了113个化合物,含黄酮类化合物72个,绿原酸及衍生物7个、香豆素类8个。这些化合物可能是黄山贡菊具备抗氧化活性的物质基础。Abstract: The antioxidant activity and chemical composition of extracts from Huangshan Gongju was studied in this work. The extractive conditions with antioxidant activity as an indicator were optimized by response surface method. The conditions were as follows:60% ethanol aqueous, the ratio of liquid to material 35:1 (g/mL), ultrasonic time of 25 min. Under this conditions, the total antioxidant capacity of the extract was (2.134±0.011) mmol/L (RSD=1.97%, n=3), which was in good agreement with the predicted value (2.010 mmol/L). The antioxidative activity of extract was analyzed by FRAP method and DPPH method. The total antioxidant capacity of the extracts (total flavonoid in equivalent of rutin was 7.88 mg/L) was equivalent to the reduction power of 2.87 mmol/L ferrous ion. The DPPH free radical scavenging rate of the extracts (total flavonoid in equivalent of rutin was 25.85 mg/L) were 94.81%. The main antioxidant components of ethanolic extracts were analyzed by HPLC-MS and 114 compounds were identified including 72 flavonoids, 7 chlorogenic acid and their derivatives and 8 coumarin derivatives. These compounds probably is the material basis of the antioxidant activity of Huangshan Gongju.
