响应面法优化棘托竹荪孢子多糖提取工艺及抗氧化活性研究

Study on the Optimization of Extraction Technology and Antioxidant Activity of Polysaccharide from Spores of Dictyophora echinovolvata by Response Surface Methodology

  • 摘要: 为了最大限度地从竹荪孢子中提取多糖,且不破坏多糖的抗氧化活性,本文考察提取时间、提取温度、料液比、pH等因素对多糖得率及多糖活性的影响,在单因素实验的基础上,采用响应面法Box-Benhnken设计,优化棘托竹荪孢子多糖提取工艺。结果表明:综合提取效率和多糖活性等因素,得到竹荪孢子多糖提取的最适工艺参数为提取温度70 ℃、提取时间120 min、pH9、料液比为1:20。采用上述最优工艺条件进行多糖提取的实验,测得实际多糖得率为9.87%,在2~10 mg/mL范围内,孢子多糖对羟基自由基清除率的IC50为4.67 mg/mL。实验结果为下一步分离、纯化及鉴定竹荪孢子活性多糖提供基础和依据。

     

    Abstract: To improve the yield and antioxidant activity of polysaccharides from the spores of Dictyophora echinovolvata,extraction conditions of polysaccharides were optimized by response surface analysis(Box-Benhnken design)based on single factor experiments including extraction time,extraction temperature,the ratio of water volume to raw material weight and pH value. The results showed that based on the extraction efficiency and activity of polysaccharides,the optimum extraction conditions were determined as follows:Extraction temperature 70 ℃,extraction time 120 min,solid-to-water ratio 1∶20,and pH9.Under these conditions,the maximum polysaccharides yield of 9.87% was obtained. The results from the antioxidant tests in vitro with polysaccharides showed that the IC50 values of scavenging against hydroxyl free radical was 4.67 mg/mL in the concentration of 2~10 mg/mL. The results provided a basis for further separation,purification and identification of polysaccharides from the spores of Dictyophora echinovolvata.
