Abstract:
Objective:To isolate, purify and screen endophytes from
Salvia miltiorrhiza seeds, which can inhibit plant pathogenic fungi and possess antioxidant and anticoagulant activities. Methods:Endophytes were isolated from seeds of
Salvia miltiorrhiza by the plate method, the endophytic fungi which had antifungal effects were isolated by the method of plate confrontation and filter paper. The DPPH method was used to measure the antioxidant activity. The anticoagulation activity of endophytic bacteria was measured in the peripheral vein of rabbit ear. Results:11 endophytic fungi and 2 endophytic bacteria were isolated from seeds of
Salvia miltiorrhiza. 9 endophytic fungi and 9 endophytic bacteria were isolated from seeds of
Salvia miltiorrhiza. Among them,a strain of endophytic fungus numbered as ZJZD-3 could restrain the pathogenic fungi No. 4 and No. 6 of
Salvia miltiorrhiza, its inhibition rate was 70.1% and 59.6%,respectively. And the ZJZD-3 had the strongest antioxidant capacity, the DPPH scavenging rate was 91.44% with 0.5 mg/mL ethanol crude extract of ZJZD-3. In addition,ZJZD-3 had extended effect on intrinsic and exogenous coagulation, especially on intrinsic coagulation. The extension rates of ZJZD-3 to rabbits TT,PT and APTT were 48.48%,12.38% and 34.44%,respectively. The ZJZD-3 was identified as
Talaromyces. Conclusions:Endophytic fungus ZJZD-3 has good ability of inhibiting plant pathogenic fungi, antioxidant and anticoagulant. It can improve new ways and methods for the development of
Salvia miltiorrhiza resources and high-quality and high-yield cultivation.
