Abstract:
In this study,Box-Behnken-double response surface optimization method combined with Matlab analysis method to be used to optimize the grape seed polyphenols(GSP). The influence of four single factors(extracted medium,solvent concentration,solid-liquid ratio,extraction time)on the content and hydroxyl radical scavenging rate of GSP were investigated. By the optimization of dual response surface combined with Matlab analysis,the optimal extraction process parameter was as follows:Acetone concentration of 78.7%,solid-liquid ratio of 1:45 g/mL,extracting time of 91.2 min. On this condition,the actual value of polyphenol extraction amount was(75.39±0.10)μg gallic acid equivalent(GAE)/mg dry weight(DW),and the hydroxyl radical scavenging rate was 70.82%±0.11%. In addition,through Matlab analysis,when the extraction time was taken as the median value(C=90 min),y

2 were both able to obtain the largest theoretical values. When acetone concentration was within the range of 76%~82%,and the closer the solid-liquid ratio was to 1:45 mL/g,the closer the values of y
1 and y
2 were to the maximum value. The results of dual response surface optimization and Matlab optimization were consistent. The combination of the two methods can not only accurately obtain the best extraction scheme,but also more intuitively find out the reasonable process parameter interval.
