HPLC-ELSD法测定长白山笃斯越桔果实中可溶性糖含量

Determination of soluble sugar in Vaccinium uliginosum fruit in Changbai Mountain by HPLC-ELSD

  • 摘要: 目的:为了更好的分析长白山笃斯越桔的食用价值,为生产加工及种质资源保存利用提供理论依据。方法:采用高效液相色谱-蒸发光散射检测器法(High performance liquid chromatography-evaporative light scattering detector,HPLC-ELSD)对2个不同居群共40份长白山笃斯越桔果实中可溶性糖含量进行测定比较。高效液相色谱条件为CAPCELL PAK NH2 UG80 S5(4.6 mm×250 mm,5 μm)色谱柱;流动相为乙腈-水(75:25,v:v);流速1.0 mL·min-1;柱温35℃;蒸发光散射检测器,漂移管温度80℃;载体为高纯N2,N2流速为1.0 L·min-1,增益值为1;进样量5 μL。结果:在选定的色谱条件下,可溶性糖分离度良好。标准曲线相关系数均在0.9994以上,回收率在102.83%~106.29%之间。长白山笃斯越桔果实主要的糖分是果糖和葡萄糖,其中果糖含量最高,海拔高的东方红林场总糖及甜度值显著(P<0.05)高于海拔低的兰家林场,DFH-11是总糖和甜度值最高的样本。结论:该方法具有操作快速、重复性好等特点,可以准确测定笃斯越桔果实中糖分类型及含量;可对笃斯越桔果实加工利用,并作为育种工作首选材料。

     

    Abstract: Objective:To analyze the edible value of Vaccinium uliginosum in Changbai Mountain and provide theoretical basis for production,processing and germplasm conservation and utilization. Methods:The soluble sugar contents in 40 V. uliginosum fruits from two different populations was determined and compared by high performance liquid chromatography-evaporative light scattering detector(HPLC-ELSD). The conditions of high performance liquid chromatography were CAPCELL PAK NH2 UG80 S5(4.6 mm×250 mm,5 μm)column,acetonitrile-water as mobile phase(75:25,v:v),flow rate 1.0 mL·min-1,column temperature 35℃ with evaporative light scattering detector(ELSD),drift tube temperature 80℃. Carrier was high purity N2,N2 flow rate was 1.0 L·min-1,gain value was 1,injection volume was 5 μL. Results:The soluble sugar was well separated under the selected chromatographic conditions. The correlation coefficients of the standard curves were over 0.9994,and the recoveries ranged from 102.83% to 106.29%. Fructose and glucose were the main sugars in V. uliginosum fruit of Changbai Mountain,and fructose content was the highest. The total sugar and sweetness values of Dongfanghong Forest Farm at high altitude were significantly(P<0.05)higher than those of Lanjia Forest Farm at low altitude,and DFH-11 was the sample with the highest total sugar and sweetness values. Conclusion:The method had the characteristics of fast operation and good repeatability,and could accurately determine the types and contents of sugar in V. uliginosum fruit. The V. uliginosum fruit can be processed and utilized as the preferred material for breeding.
