皂荚多糖超声波提取工艺优化及体外抗氧化活性研究

Optimization of Ultrasonic Assisted Extraction and Antioxidant Activity of Polysaccharide from Gleditsia japonic Miq.

  • 摘要: 以皂荚多糖为研究对象,在单因素实验的基础上,采用响应面法对皂荚多糖的超声波提取工艺进行优化,并对多糖进行体外抗氧化活性的研究。结果表明:最佳提取工艺条件为:提取温度50 ℃,液料比35:1 (mL/g),提取时间30 min,超声功率285 W,提取3次,多糖得率为30.65%±0.25%。抗氧化实验表明,皂荚多糖具有一定的抗氧化能力,其对超氧阴离子自由基、DPPH自由基、羟自由基清除作用的IC50分别为:10.3、6.9、1.5 mg/mL。

     

    Abstract: The polysaccharide in Gleditsia japonic Miq. was researched. On the basis of single factor experiment,response surface methodology was used to optimize the ultrasonic extraction process of Gleditsia japonic Miq.,and the antioxidant activity of polysaccharides in vitro was studied. The results showed that the optimum technology conditions were as follows:Extraction temperature 50 ℃,liquid to material ratio 35:1 (mL/g),extraction time 30 min,ultrasonic power 285 W,performed three times,under these conditions,the yield of polysaccharide was 30.65%±0.25%. Antioxidant activity test showed that the polysaccharide in Gleditsia japonic Miq.had certain antioxidant activity,and the IC50 of radical scavenging capacity on O2-·,DPPH· and ·OH were 10.3,6.9,1.5 mg/mL,respectively.
