舞花姜根总皂苷的提取工艺及其抗氧化活性

Study on extraction process and antioxidative activity of total saponins from Globba racemosa Smith roots

  • 摘要: 研究舞花姜根中总皂苷的超声提取最佳工艺条件,并评价提取总皂苷的抗氧化活性。在单因素实验基础上,采用3因素3水平响应面实验优化超声提取条件。结果表明:舞花姜根总皂苷的最佳提取工艺参数为:料液比1:30 (g/mL),样品粒度40目,超声提取时间51 min。按此工艺对不同产地舞花姜根总皂苷进行提取,发现务川县和三都县的总皂苷含量相对较高且稳定,含量分别为16.88 mg/g和16.92 mg/g。利用DPPH、ABTS进行抗氧化分析,得出舞花姜根提取物浓度与抗氧化活性呈正比;不同产地的舞花姜根总皂苷提取物对自由基清除率不同;且清除率强弱与舞花姜根中提取的总皂苷含量呈正相关,验证了其抗氧化能力与总皂苷含量有关。

     

    Abstract: The purpose was to explore the optimal ultrasonic extraction conditions of total saponins from Globba racemosa Smith roots, and evaluate the antioxidant activity.On the basis of single factor experiment, 3 factors and 3 level response surface test was adopted. The results showed that the optimum extraction parameters of the total saponins in the Globba racemosa Smith roots were the ratio of liquid to material was 1:30 (g/mL), the particle sizes of sample were 40 mesh and the ultrasonic extraction time was 51 min. According to this process, the total saponins in the Globba racemosa Smith roots were extracted in different habitats, the content of total saponins in Wuchuancounty and Sanducounty were relatively high and stable, with content of 16.88 mg/g and 16.92 mg/g respectively. The antioxidant analysis of using DPPH and ABTS showed that the concentration of extracting in the Globba racemosa Smith roots was proportional to the antioxidant activity. The free radical scavenging rate of the total saponins extract of the Globba racemosa Smith roots was different in different habitats, and the intensity of the free radical scavenging rate was positively correlated with the total saponins content extracted from Globba racemosa Smith roots. It is verified that the antioxidant ability was related to the total saponins content.
