适配体在食品中有害小分子检测领域应用的研究进展

Advances in aptamer for the detection of toxic small molecules in food

  • 摘要: 适配体是与靶标具有高亲和力及选择性结合能力的单链寡核苷酸,可通过指数富集的配基系统进化技术(Systematic Evolution of Ligands by Exponential Enrichment,SELEX)筛选得到。SELEX技术可选择的靶分子范围十分广泛,包含蛋白质、多肽、核苷酸、染料、氨基酸、金属离子、生物毒素等;经SELEX技术筛选出的适配体具有特异性强的特点;同时该技术具有周期短的优点;筛选用的文库化学合成即可,避免了动物实验。食品中有害小分子主要包括真菌毒素、农药、兽药、金属子、违禁添加物等,主要存在于粮食、谷物、饲料、各类食品、水中。这些有害小分子通过食物链进入人及动物体内,严重威胁到其健康。目前已有不少文献报道了以适配体作为识别元件,用于食品安全、环境检测及生物医药等领域的多种高亲和力及特异性的传感器和检测方法的建立。本文主要介绍了近年小分子适配体在食品安全检测中应用的研究进展。 

     

    Abstract: Aptamers are single-stranded oligonucleotides with high affinity and selective binding to the target and can be screened by SELEX techniques.SELEX technology can choose a wide range of target molecules, including proteins, peptides, nucleotides, dyes, amino acids, metal ions, biological toxins.Aptamer selected by SELEX has specific characteristics.With the advantages of short cycle, the library can be chemical synthesis to avoid the animal experiments.Food harmful small molecules mainly include mycotoxins, pesticides, veterinary drugs, metal, prohibited additives, mainly in food, grain, feed, all kinds of food, water.These harmful small molecules through the food chain into the human and animal body and create a serious threat to its health.A variety of high affinity and specific sensors and detection methods have been reported in previous, aptamers are used as identification element for food safety, environmental testing and biomedicine.Unlike previous reports, this paper mainly foucusses on the advances in aptamer for the detection of toxic small molecules in food.
