合浦珠母贝糖胺聚糖的分离纯化及理化性质分析

Purification and physicochemical characteristics of glycosaminoglycan from Pinctada martensii

  • 摘要: 研究合浦珠母贝糖胺聚糖粗品经分离纯化得到的精制品的理化性质,通过琼脂糖凝胶电泳鉴定样品分离情况,并对样品进行红外分析。经枯草杆菌蛋白酶与胰蛋白酶双酶酶解提取糖胺聚糖粗制品后,将粗制品利用DEAE-52离子交换柱进行纯化,纯化时分别用0.5 mol/L Na Ac(p H=6.0)和1.5 mol/L Na Cl进行洗脱,得到精制品GAG1和GAG2。利用苯酚-硫酸法、Bradford法、明胶-氯化钡法、Orcinol法、Wagner法、红外光谱法分析GAG1和GAG2的理化性质。通过各实验结果,可以初步猜测GAG1为肝素钠,GAG2为硫酸软骨素。 

     

    Abstract: This paper studied on purification and physicochemical characteristics of glycosaminoglycan from Pinctada martensii.The separation of the samples was identified by agarose gel electrophoresis and analyzed by infrared spectroscopy. After glycosaminoglycan was achieved from the whole viscera of Pinctada martensii via enzymatic hydrolysis by pancreatin and neutral protease of Bacillus subtilis,the crude glycosaminoglycan was purified by DEAE-52 ion exchange column with 0.5 mol / L Na Ac( p H = 6.0) and 1.5 mol / L Na Cl,then the purified product of glycosaminoglycan( GAG1 and GAG2) were obtained. The physicochemical properties of the purified product were analyzed by the method of phenol- sulfuric acid,Bradford,Orcinol,Wagner,Infrared spectroscopy.The results showed that GAG1 was heparin sodium and GAG2 was chondroitin sulfate.
