响应面法优化重组运动发酵单胞菌极高密度乙醇发酵工艺

Optimization of very high ethanol fermentation conditions of the recombinant Zymomonas mobilis by response surface method

  • 摘要: 采用基因工程技术,成功地构建了在无需氨基酸和维生素的条件下,能高效利用葡萄糖、木糖、阿拉伯糖和甘露糖发酵生成乙醇的重组菌Z.mobilis HYMX。为了提高发酵乙醇产量,利用响应面法优化了重组菌极高密度发酵的工艺条件。结果表明,最佳发酵条件为:葡萄糖浓度305 g/L,温度34℃,p H6.5,尿素浓度3 g/L,接种量10%,发酵时间60 h。在此条件下重组单胞菌的乙醇产量达到153.1 g/L,比优化前提高了8.5%,比原型菌产量提高了105.14%。与工业酵母菌株PE-2相比重组菌节省了约12%的葡萄糖,节省了37.5%的时间。 

     

    Abstract: The recombinant bacteria Z.mobilis HYMX which can utilize glucose,xylose,arabinose and mannose to produce ethanol in the culture medium without amino acids and vitamins,was constructed by genetic engineering technology.The fermentation conditions of the HYMX strain under very high gravity ethanol fermentation conditions were optimized by the response surface methodology. The results showed that the optimum conditions were:glucose concentration 305 g / L,temperature 34 ℃,p H6.5,urea concentration 3 g / L,inoculation amount 10%,fermentation time 60 h.Under the optimized conditions,the ethanol yield of the recombinant strain was 153.1 g / L,8.5% higher than the non- optimized one,105.14% higher than the prototype strain. Compared with the industrial yeast strain PE-2,the glucose saving was more than 12%,and the fermentation time saving was more than 37.5%.
