Abstract:
One β-glucosidase producing strain was selected from various soil samples and dyeing wastewater by using esculin agar plate screening method with the aim to study and develop alkaline microbial origin β-glucosidase resources,and the microbiology identification,fermentation properties and enzyme characterization of the isolated strain were further studied. The isolated strain was identified as Bacillus sp.,and the optimum growth and enzyme production temperatures was between 35 ~37 ℃,and the optimum growth and enzyme production p H were at 9.5 and 9.0,respectively. The optimum temperature and p H of the produced enzyme was 50 ℃ and 9.5,respectively,and the enzyme was stable at temperatures below 50 ℃ and in a p H range of9.0~10.0. Mental ions,such as Cu~(2+),Fe~(2+),Ca~(2+) and Mn~(2+) showed some inhibition to the enzyme,Mg~(2+),Zn~(2+) and Co~(2+)have no significantly effect on the catalytic activity of the enzyme. The results obtained in the study were meaningful for microbial resources and information consolidation of alkaline β-glucosidase producing strain.
