扁杏仁肽对血管紧张素转化酶的抑制作用研究

摘要: 采用碱溶酸沉法从陕北扁杏仁制得杏仁总蛋白,并采用Osborne法分级制备清蛋白、醇溶蛋白、球蛋白和谷蛋白;用碱性蛋白酶Alcalase水解,超滤分离,制备不同分子量总蛋白杏仁肽和分级蛋白杏仁肽;利用高效液相色谱法测定水解物,系统研究杏仁肽对血管紧张素转化酶(ACE)的抑制活性。结果表明,不同分子量的总蛋白杏仁肽均具有ACE抑制活性,且肽段分子量越小,抑制活性越强;分级蛋白杏仁肽中,谷蛋白肽的抑制活性最强,分子量低于1 ku的谷蛋白肽的IC50值为(96.21±0.06)μg/m L。Abstract: The sweet almond protein was prepared with the method of alkali extraction and acid precipitation,and then fractionated as albumin,glindin,globulin and glutelin. The proteins were hydrolyzed as peptides with Alcalase protease and the hydrolysates were filtered through polyethersulfone membrane with different molecular weight cutoffs. The angiotensin-converting enzyme(ACE) inhibitory activities of different molecular weight of almond peptides were measured. The results showed that different molecular weight peptides had different inhibitory activities and the activity increased with the molecular weight reduced. The results also showed that the activity had obviously difference for peptides derived from different proteins,glutelin peptides were highest activity among four proteins and the IC50 of ACE inhibitory activity was(96.21±0.06) μg/m L for glutelin peptide with molecular weight below 1 ku.
