鳕鱼骨胶原肽的制备及其对成骨样MG-63细胞作用机制的研究

摘要: 以水产加工下脚料鳕鱼骨为原料,对其进行基本组成分析与组织结构观察。从鳕鱼骨中提取胶原蛋白并酶解制得不同分子量段的胶原肽。探讨不同分子量胶原肽组分对MG-63细胞碱性磷酸酶(ALP)活性及相关因子骨形态发生蛋白(BMP-2)、骨保护素(OPG)与核因子-κB受体活化因子配基(RANKL)m RNA表达水平的影响。结果表明:鳕鱼骨中蛋白质含量较高(14.5%),Van Gieson染色发现鳕鱼骨中含有大量的胶原蛋白纤维。鳕鱼骨胶原肽能够提高细胞ALP活性,同时显著上调细胞BMP-2 m RNA的表达水平;鳕鱼骨胶原肽可有效提高细胞OPG/RANKL m RNA的相对表达量,以间接对破骨细胞的成熟及活化进行调控。鳕鱼骨可作为一种优良的天然蛋白源,鳕鱼骨胶原肽能够作为一种有效的抗骨质疏松营养强化剂加以开发。Abstract: The cod bone collagen hydrolysates were prepared by commercial protease. Utilizing cultured MG-63 cells, the osteogenic effect of collagen hydrolysates with different molecular weights was studied in vitro.Compositional analyses and histological observation was performed. Cod bone collagen hydrolysates with different molecular weights were obtained. The cultured MG-63 cells were used to investigate the osteogenic effect by detecting the ALP activity and the m RNA expression of Bone morphogenetic protein( BMP- 2),Osteoprotegerin(OPG) and Receptor activator of nuclear factor-κB ligand(RANKL. The proximate analysis of fish bone exhibited that protein content in bone reached 14.5%. Large amounts of collagen fibrils in fish bone were observed using Van Gieson staining technology. Cod bone collagen hydrolysates could increase the ALP activity and BMP-2 m RNA expression level,meanwhile remarkably increase the ratio of OPG/RANKL m RNA expression in order to indirectly influence the osteoclast differentiation and maturation. Cod bone was a good collagen resource,the collagen hydrolysates extracted from cod bone exhibited osteogenic effect and could be used as a natural anti-osteoporotic supplement.
