HPLC-DAD法测定通贤柚叶中两种活性黄酮的含量

Determination of two flavonoids in Tongxian grapefruit leaves by HPLC-DAD

  • 摘要: 目的:建立HPLC-DAD测定通贤柚叶中柚皮苷与野漆树苷的方法。方法:色谱柱为Sepax Amethyst C18-H(250×4.60mm,5μm),流动相为乙腈-0.1%甲酸梯度洗脱,流速1.0m L/min,检测波长为283nm(柚皮苷),337nm(野漆树苷),柱温25℃,进样量20μL。结果:柚皮苷与野漆树苷色谱峰分离良好。二者分别在0.06530.6526mg/m L与0.03500.3505mg/m L范围内与峰面积呈良好的线性关系。平均加样回收率分别为98.68%与98.47%。结论:所建立的含量测定方法可用于通贤柚叶的质量控制。采收季节与叶龄对柚叶中两种黄酮类物质的含量有较明显的影响。 

     

    Abstract: Objective: To develop a HPLC-DAD method to determine the content of naringin and rhoifolin in Tongxian grapefruit leaves.Methods: HPLC was performed on a Sepax Amethyst C18-H analytical column( 250 mm × 4.60 mm,5μm) with gradient elution using acetonitrile and 0.1% aqueous formic acid as the mobile phase at a flow rate of1.0m L/min. The detection wavelength was 283 nm for naringin,337 nm for rhoifolin. The column temperature was25℃ and the injection volume was 20μL. Results: The chromatographic peaks of naringin and rhoifolin could be separated well in the above cases.The linear ranges for the two flavonoids were 0.0653~0.6526 mg/m L and 0.0350~0.3505 mg/m L,respectively. The average recoveries were 98.68% and 98.47%,respectively.Conclusion: The established method could be used for the quality control of Tongxian grapefruit leaves.Harvest season and leaves age affect the content of two flavonoids significantly.
