Determination of antiviral drugs residue in chicken by UPLC-MS/MS

  • 摘要: 采用超高效液相色谱质谱(UPLC-MS/MS)法建立了鸡肉中利巴韦林和金刚烷胺等抗病毒类药物残留量的检测方法。鸡肉样品均质后,分别用乙腈-1%三氯乙酸溶液和甲醇-1%三氯乙酸溶液提取,提取液经固相萃取小柱净化,采用多反应监测模式进行检测,外标法定量。添加浓度为0.12.0μg/kg时,抗病毒类药物的回收率为66.7%85.6%,相对标准偏差为3.1%8.5%。方法简便快捷,精密度高,重现性好,可用于鸡肉中抗病毒类药物的测定。 
    Abstract: An ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) method was established to determine residue of ribavirin and amantadine in chicken. Homogenized samples were extracted separately with a mixture of acetonitrile-1% trichloroacetic acid aqueous solution and a mixture of methanol-1% trichloroacetic acid aqueous solution, and then cleaned by solid phase extraction (SPE) . UPLC-MS/MS detection was performed using multiple reaction monitoring module. The quantification was obtained by utilizing the external standard method. Recoveries of metabolites of antiviral drugs ranged from 66.7% to 85.6% at fortified levels of 0.1 ~2.0μg/kg. The RSDs of the method were in the rang of 3.1% ~8.5%. The method was simple, effective and accurate. It could be applied to determine antiviral drugs residue in chicken.
