反相高效液相色谱法测定泡萝卜中的有机酸
Analysis of organic acids in pickled radish by reverse phase high performance liquid chromatography

摘要: 本研究建立了反相高效液相色谱法同时分析泡萝卜中8种有机酸的方法。最佳色谱条件为AQ C18色谱柱(5μm,120A,4.6mm×150mm),4%甲醇-96%的0.05mol/L KH2PO4(H3PO4调节pH为2.9)作为流动相,流速0.6mL/min,柱温27℃,进样量10μL,检测波长210nm;最佳有机酸提取方法为超纯水提取法。利用上述条件进行测定,能够有效的对泡萝卜中的8种有机酸进行提取、分离和测定,回收率为90.59%103.85%,RSD为0.34%2.18%。Abstract: A method for simultaneous determination of eight organic acids in pickled radish by reverse phase high performance liquid chromatography was established. YMC- Pack ODS- AQ C18column ( 5μm, 120A, 4.6mm ×150mm) was used at 27℃ and mobile phase was 4% CH3OH- 0.05 mol /L KH2PO4 ( pH2.9) with flow rate of0.6mL /min. Detection wavelength was 210nm.The extraction with ultrapure water was better.With these conditions, eight organic acids in pickled radish could be effectively separated and detected.The recoveries were 90.59% to103.85%, and the relative standard deviations were 0.34% to 2.18%.
