优化蚕蛹多糖的提取方法及脱色工艺的研究

Study on the process optimization of extraction of polysaccharide of silkworm pupa and decolorization

  • 摘要: 目的:优化蚕蛹多糖的提取工艺及对蚕蛹多糖的脱色条件的研究。方法:采用正交实验方法,以蚕蛹多糖的得率为指标,优化微波法对蚕蛹多糖的提取工艺。研究离子交换树脂(D202、D113)和大孔吸附树脂(HZ-841、HZ-806、HZ-803)对蚕蛹多糖的脱色效果。通过正交实验确定蚕蛹多糖脱色的最佳条件。结果:传统工艺提取最佳条件:90℃提取3.5h,固液比1∶25g/mL,蚕蛹多糖的得率为3.95%;微波提取蚕蛹多糖的最佳工艺条件:微波功率600W,提取时间9min,固液比1∶25g/mL,蚕蛹多糖的得率为4.49%。蚕蛹多糖大孔树脂脱色的最佳条件为HZ-803树脂、pH4.0、温度45℃。结论:优化之后的微波提取法提取蚕蛹多糖的得率有显著的提高,且工艺简便、合理、可行,在大孔树脂静态吸附最佳脱色的条件下脱色率、多糖保留率和蛋白质去除率分别为88.56%、62.15%、92.21%,在大孔树脂动态吸附最佳脱色的条件下脱色率、多糖保留率和蛋白质去除率分别为89.43%、65.58%、90.56%。 

     

    Abstract: Objective:Optimize the extraction technic of polysaccharide of silkworm pupa and study the condition of decolorizes the polysaccharide. Methods:By means of orthogonal experiment, optimize the extraction technic of polysaccharide used microwave method according to the yield index of polysaccharide. Explored the decolorizing effect of polysaccharide by using the ion exchange resin (D202, D113) and macroporous adsorptive resin (HZ-841, HZ-806, HZ-803) and found the optimum conditions of the decolorization of polysaccharide by orthogonal experiment. Results:The optimum conditions of the traditional extraction technic were extraction 3.5h at 90℃、ratio of solid to liquid 1∶25, the yield index of polysaccharide was 3.95%;the optimum conditions of the microwave extraction of polysaccharide were microwave power 600W, extraction time 9min, ratio of solid to liquid 1 ∶25, the yield index of polysaccharide was 4.49%. The optimum conditions of the decolorization of polysaccharide was HZ-807 Resin、pH4.0、temperature 45℃. Conclusion:It was significantly increased of the yield index of polysaccharide by using the optimized microwave extraction method, and the process was simple、reasonable and feasible. In the most appropriate experimental condition, the decolorization rate of polysaccharide of silkworm pupa, polysaccharide retention rate and protein removal rate was 88.56%, 62.15%, 92.21% with macroporous resin by static absorbing test. In the most appropriate experimental condition, the decolorization rate of polysaccharide of silkworm pupa, polysaccharide retention rate and protein removal rate was 89.43%, 65.58%, 90.56% with macroporous resin by dynamic test test.
