利用TLC和HPLC结合的方法定性检测尿液中的雌马酚

摘要: 雌马酚是人体肠道细菌分解大豆苷和大豆苷元的最终代谢产物之一,采用薄层色谱法(TLC)分离纯化尿液样品中的雌马酚后再进行高效液相色谱法(HPLC)定性检测,可有效减少染料木苷元的干扰,解决了仅依靠HPLC无法实现雌马酚有效分离的问题。以氯仿-甲醇(24∶1,v/v)作为TLC展开剂,染料木苷元的Rf值为0.29,雌马酚的Rf值为0.40。本方法具有准确度高,干扰度低等优点,为定性检测尿液中雌马酚提供了新的方法。Abstract: Equol was one of metabolites of daidzin and daidzein by intestinal bacteria decomposition. The disturbance from genistein to equol was reduced by using thin-layer chromatography (TLC) followed by high performance liquid chromatography ( HPLC) . This method solved the problem that equol could hardly be separated effectively by HPLC. Developed in a TLC solvent system of chloroform∶methanol (24∶1) , equol and genistein were observed on Rf value of 0.40 and 0.29, respectively. This new qualitative method for the analysis of urinary equol was more accurate and less disturbed than using only HPLC analysis.
