Optimization of process conditions for the extraction of chondroitin sulfate by response surface methodology

摘要: 采用鸡胸软骨为原料,以硫酸软骨素得率为指标,对超声波辅助碱-酶法提取硫酸软骨素的工艺条件进行研究。首先,通过单因素实验确定了各因素的范围,再应用Box-Behnken中心组合实验设计建立二次多项数学模型,进行响应面分析。结果表明,超声波辅助碱-酶法提取鸡软骨中硫酸软骨素的最佳工艺条件为:碱浓度4%,酶添加量1.4g/L,料液比1000∶5923(g/mL),数据结果显著,硫酸软骨素得率为23.94%,通过紫外光谱检测确定提取物质为硫酸软骨素。Abstract: The chicken sternal cartilage was used as the raw material and the chondroitin sulfate extraction yield as the target to withdraw the method by ultrasonic assisted alkali-enzymatic. Firstly, the ranges of all affecting factors were determined by single factor experiment and Box-Behnken center combination experimental design used to set up two times multiterm mathematical model. The result indicated that the data results significantly and the optimum conditions of ultrasonic assisted alkali-enzymatic were obtained as follows :the alkali concentration was 4%, enzyme dosage was 1.4g/L, ratio material to solvent was 1000 ∶5923 (g/mL) , and the chondroitin sulfate extraction yield was 23.94%. Determined the extraction of substances through the UV spectrum detection of chondroitin sulfate.
