分光光度法测定牦牛不同脏器中左旋肉碱方法优化及含量分析

Optimization the L-carnitine determination by spectrophotometric method and comparison of content in the yak organs

  • 摘要: 对牦牛脏器中左旋肉碱的分光光度法测定进行优化,并对其脏器中左旋肉碱含量进行分析。在单因素实验的基础上,以加酶反应时间、缓冲体系pH、酶反应温度为自变量,以吸光度为响应值,进行响应面分析。确定最优工艺条件为:酶反应温度37℃、缓冲体系pH7.6、加酶反应时间16min,在此条件下,左旋肉碱吸光度值为0.321。通过对牦牛心、肝、肺、肾中左旋肉碱测定值的分析发现,肝脏中左旋肉碱含量最高,为28.35mg/kg;肺脏中左旋肉碱含量为22.75mg/kg;心脏中含左旋肉碱14.61mg/kg;肾脏中左旋肉碱的含量最低,为5.22mg/kg。 

     

    Abstract: The spectrophotometric determination method about L-carnitine in organs of yak was optimized and the content of L-carnitine in the yak organs was analyzed in this paper. Based on the single factor experiment, the response surface methodology was used to select the enzyme reaction time, pH of buffer system and enzyme reaction temperature. These three was shown as variables, and absorbance as response value. The optimum conditions were as follows:the enzyme reaction temperature was 37℃, pH was 7.6 and enzyme reaction time was 16 minutes. Under this condition, the absorbance of L-carnitine was 0.321. The measured value of yak heart, liver, lung and kidney was analyzed under the optimum condition. And the result was shown that the highest L-carnitine yield of 28.35mg/kg was achieved from liver, then was lung of 22.75mg/kg and heart of 14.61mg/kg, kidney had the lowest content of 5.22mg/kg.
