Determination of L-citrulline in watermelon by high performance liquid chromatography with PITC precolumn derivation

摘要: 建立了RP-HPLC测定西瓜中L-瓜氨酸的方法。西瓜试样采用异硫氰酸苯酯(PITC)衍生后用液相色谱仪在245nm下进行测定,结果表明,L-瓜氨酸质量浓度在4.26213.2μg/mL范围时,其质量浓度与峰面积呈良好的线性关系(R=0.9998)。L-瓜氨酸的最低检出限为1.1μg/mL,相关系数为0.9998,平均回收率为98.1%。参试西瓜试样中L-瓜氨酸含量在1.132.71mg/g之间,该方法前处理简单,灵敏、快速、可靠,适合用于西瓜样品中L-瓜氨酸含量的定量测定。Abstract: A method of RP-HPLC with phenylisothiocyanate (PITC) precolumn derivatization was established for the determination of L-citrulline in watermelon. The calibration curves of L-citrulline were linear over the range of 4.26213.2μg/mL (R=0.9998) and the recovery rate was 98.1%. The content ranges of L-citrulline in watermelon were 1.132.71mg/g. The detection limit of the method was 1.1μg/mL. The method descried in this paper was fast, sensitive and reliable and it was most suitable for quantitative determination of L-citrulline in watermelon.
