  • 摘要: 采用可生物降解的壳聚糖为原料,与多聚磷酸钠和环氧氯丙烷通过离子和共价作用形成交联壳聚糖,研究了交联壳聚糖对牛血清蛋白(BSA)的吸附和释放行为。考察了BSA初始浓度、交联壳聚糖的用量、pH等对吸附效果的影响;通过正交实验设计优化了反应条件;并进行了吸附动力学的研究。结果显示:在最优的条件下,交联壳聚糖对牛血清蛋白的最大吸附率为88.7%;吸附数据可用伪二级动力学方程来描述,符合Freundlich等温吸附模型;负载牛血清蛋白的交联壳聚糖易被氯化钠洗脱,并可重复利用。 

     

    Abstract: Using biodegradable chitosan as material, the cross-linked chitosan beads were prepared by crosslinking chitosan with both sodium polyphosphate and epichlorohydrin via ionic and covalent bonds. The adsorption and desorption of bovine serum albumin (BSA) onto cross-linked chitosan were investigated. The effects of initial concentration of BSA, cross-linked chitosan dosage, pH on the adsorption were evaluated, and the adsorption conditions were optimized by orthogonal experiment. The analysis of kinetics for adsorption was preformed. The results showed that the maximum adsorption rate for BSA was 88. 7% under the optimum conditions. The adsorption phenomena appeared to follow the pseudo-second order kinetics equation and Freundlich adsorption equation. The cross-linked chitosan carrying BSA could be reused after elution easily by NaCl.
