Abstract:
To optimize the purification technology and parameters of macroporous resins to total flavonoids in Lycium barbarum L.from Qaidam Basin.The HPD400 was optimized from 11 types of macroporous resins by comparing their static adsorption and desorption performances with the total flavonoids in Lycium barbarum L.from Qaidam Basin.Single-factor tests and response surface method (RSM) were used for optimizing the purification technology.The optimal purification condition as follows:sample loading amount were 16.0mL, pH4.0, and the velocity of sample solution 1.0mL/min, rinsing the column with 3BV deionized water after completely adsorption, and then 80% ethanol as eluent, the volume 23.0mL and the velocity 1.0mL/min.Under these optimal parameters, total flavonoids recovery rate were up to 89.92%, and the content of total flavonoids purified by HPD400 macroporous resin increased from 10.80% to 27.62%.
