鸭小肠抗菌肽的分离纯化及抗菌活性研究

摘要: 为了探究鸭肠道抗菌肽的高效提纯方法和抗菌活性,本研究将新鲜鸭肠黏膜破碎,高温处理,乙酸浸提后,再经超滤和Sephadex G50凝胶过滤层析,收集各组分并用琼脂糖弥散法检测抑菌活性。同时,采用TricineSDS-PAGE测定其主要抗菌肽的分子量。结果表明:采用"改良乙酸浸提法"可有效提取鸭肠道抗菌肽;鸭肠道抗菌肽的分子量范围为5ku以上,其中主要抗菌肽的分子量约为6.5ku,该抗菌肽对大肠杆菌、金黄色葡萄球菌、沙门氏菌均表现出较强的抗菌活性。Abstract: To explore the highly-efficient method of purifying antibacterial peptides from the small intestine of duck and antibacterial activities of the antibacterial peptides, the mucous membrane of small intestine had been cracked, heated and lixiviated from 5% acetic acid, and then after ultra-filtration and Sephadex G50 gel filtration chromatography, collected to detect bacteriostatic activity by agarose dispersion method.The samples were also analyzed by Tricine SDS-PAGE dying to determine the molecular weight of the main antibacterial peptides.The results showed that the improved acetic acid purifying method was useful.The bound of molecule weight was 5ku upwards, and the main molecule weight was about 6.5ku.It was strongly against all the bacteria strains mentioned above, such as Escherichia coli, Salmonella and Staphyloccocus aureus.
