Anti-photoaging Effect and Mechanism of Tomato and Aronia melanocarpa Extract

  • In order to investigate the anti-photoaging effect and mechanism of tomato and aronia extracts, the effects of tomato extract and aronia extract on apoptosis, reactive oxygen species (ROS) and prostaglandin E2 (PGE2) were studied in UVB-irradiated HaCaT cells. Hematoxylin-eosin (HE) staining was used to evaluate the anti-photoaging effect of tomato and aronia complex (LA) on skin photoaging in UVB-irradiated ICR mice. The activity of superoxide dismutase (SOD), the level of malondialdehyde (MDA) and the expression of pro-inflammatory factors interleukin-1β (IL-1β), interleukin-6 (IL-6), cyclooxygenase-2 (COX-2) and tumor necrosis factor-α (TNF-α) were detected. The results showed that tomato extract and aronia extract were not cytotoxic to HaCaT cells. In the photoaging model of HaCaT cells, compared with the model group, the number of apoptotic cells in 100 μg/mL tomato extract group and 30μg/mL aronia extract group were significantly decreased (P<0.05). Aronia extract (100 μg/mL) significantly decreased ROS level (P<0.01). At 100 μg/mL, PEG2 was significantly decreased in both tomato and aronia extract groups (P<0.05). Compared with the model group, LA treatment group resulted in a significant reduction in dermal thickness (P<0.01) and MDA levels (P<0.01), and a significant increase in SOD activity (P<0.01) in skin tissue, while the proinflammatory factors IL-6, COX-2 and TNF-α were all significantly decreased (P<0.05). Tomato and aronia complex can improve oxidative stress and inflammation induced by UVB, and has the effect of anti-photoaging.
