1H-NMR Fingerprint of Hengshan Astragalus membranaceus

  • Objective: To establish a hydrogen nuclear magnetic resonance (1H-NMR) fingerprint of Hengshan Astragalus membranaceus, and provide a basis for its quality control. Method: The chemical components were extracted by ultrasonic assisted enzymatic method. Using deuterated chloroform as the extraction solvent, a standard pre-saturated pulse sequence was used to suppress water peaks at 25 ℃ to establish 1H-NMR fingerprints of 10 batches of A. membranaceus from different sources. The signal attribution was assigned by the qualitative standard addition test, database comparison and related literature comparison. The correlation coefficient method was used to calculate the similarity of 1H-NMR fingerprints, and the repeatability, precision and stability were investigated. Principal component analysis based on SIMCA was used to evaluate the quality of medicinal materials. Results: 24 compounds were detected and analyzed from Hengshan A. membranaceus, including triterpene saponins, isoflavones, amino acids, organic acids, sugars and so on. The similarities of fingerprints were higher than 0.9 with an average of 0.975. Compared with Mongolian Astragalus membranaceus and Gansu Astragalus membranaceus, the content of isoflavones in Hengshan Astragalus membranaceus was higher than both. PCA results showed that the cumulative contribution rate of the first three principal components was 89.8%, which was basically consistent with the similarity evaluation results. Conclusion: The established 1H-NMR method had the advantages of quickness and simplicity, which could provide reference for the quality control of Hengshan A. membranaceus.
