Effects of Different Extraction Methods on the Content of Phenolic Compounds in the Aralia elata(Miq.) Seem.

  • For the purpose of promoting the extraction rate of phenolic compounds from Aralia elata (Miq.) Seem., five methods including solvent extraction, solvent reflux extraction, ultrasonic-assisted extraction, microwave-assisted extraction and combined-enzyme extraction were respectively applied. After comparing the extraction rate of the five methods, then the most suitable method was obtained and orthogonal experiment was carried out to determine its optimum parameters. Result showed that the combined-enzyme extraction is more suitable for the extraction of phenolic compounds compared with other methods. According to the orthogonal experiment, its optimum extraction parameters were as follows: Enzyme dosage (composed of pectinase and various carbohydrate enzymes) 120 µL/g (enzyme amount/ sample weight), enzymatic hydrolysis temperature 50 ℃, pH6, enzymatic hydrolysis time 1.5 hours, ethanol concentration of 50%. Finally, under the aforementioned optimum technology conditions, the extraction rate of total phenols from the powder of Aralia elata (Mip.) seem. was 7.24% and total flavonoids was 3.17%. In conclusion, through the orthogonal experiment and combined-enzyme extraction, phenolic compounds from Aralia elata obtained the ideal extraction effect which could be studied for further purification and identification experiment.
